چکیده

The design of well-characterized, adaptable, blood-perfused tissue models has become a central objective of the emerging field of tumor engineering, as physiologically accurate in vitro vascular models can provide new avenues for modulation of the tumor microenvironment and preclinical evaluation of the therapeutic potential of new treatments. The characterization of fluid parameters in perfused tissue models is a critical step towards understanding and manipulation of the tumor microenvironment. The recent development of x-ray phase contrast particle image velocimetry has enabled measurement of blood flow in simple systems. This study presents high-resolution blood velocity measurements in a three-dimensional collagen microvessel bioreactor using synchrotron xray illumination. These results provide the groundwork for future flow measurements in a fully functioning blood-perfused tumor model.

Flow Measurements in a Blood-Perfused Collagen Vessel Using X-Ray Micro-Particle Image Velocimetry

Blood-perfused tissue models are joining the emerging field of tumor engineering because they provide new avenues for modulation of the tumor microenvironment and preclinical evaluation of the therapeutic potential of new treatments. The characterization of fluid flow parameters in such in-vitro perfused tissue models is a critical step towards better understanding and manipulating the tumor mi...

Hydrodynamic Characterization of an Arterial Flow Bioreactor

In vitro arterial flow bioreactor systems have been used in tissue engineering as models to investigate the response of endothelial cells to shear. However, the degree to which such models can reproduce physiological flow has not been quantified. Furthermore, previous works have assumed a Poiseuille flow profile for the calculation of wall shear stresses (WSS) experienced by the endothelial lay...

X-ray PIV measurement of blood flow in deep vessels of a rat: An in vivo feasibility study

X-ray PIV measurement is a noninvasive approach to measure opaque blood flows. However, it is not easy to measure real pulsatile blood flows in the blood vessels located at deep position of the body, because the surrounding tissues significantly attenuate the contrast of X-ray images. This study investigated the effect of surrounding tissues on X-ray beam attenuation by measuring the velocity f...

In vivo measurement of hemodynamic information in stenosed rat blood vessels using X-ray PIV

Measurements of the hemodynamic information of blood flows, especially wall shear stress (WSS), in animal models with circulatory vascular diseases (CVDs) are important to understand the pathological mechanism of CVDs. In this study, X-ray particle image velocimetry (PIV) with high spatial resolution was applied to obtain velocity field information in stenosed blood vessels with high WSS. 3D cl...

Velocity Measurement of Blood Flow in a Microtube Using Micro PIV System

Biophysical behavior of blood in the vasculature is of importance in microcirculation researches. Especially, rheological properties of blood flow have been studied using in vivo and in vitro experiments. This paper describes flow characteristics of in vitro blood flow. In order to obtain velocity distributions with high spatial resolution, a micro PIV technique, which consists of a fluorescent...
